Composition
Each 12 mL PENTOSAN GOLD INJECTION vial contains:
1,500 mg Pentosan Polysulfate (125 mg/mL)
2,400 mg N-Acetyl Glucosamine (200 mg/mL)

Features and Benefits
Active ingredients Pentosan Polysulfate, Glucosamine and Hyaluronic Acid are well documented and utilised individually in the treatment of joint disease yet their actions are closely linked. The logical combination of all 3 products for single intravenous injection aims to enhance the horse's response to treatment.

Pentosan Polysulfate
Pentosan polysulfate (PPS) produces anti-inflammatory, anti-arthritic and chondroprotective effects via 5 main actions:
1. Stimulation of cartilage cells (chondrocytes) to increase the synthesis of proteoglycans;
2. Stimulation of hyaluronic acid (HA) production by synovial cells to increase the volume and viscosity of synovial fluid;
3. Direct anti-inflammatory activity within and around the joint by inhibiting the arachidonic acid cascade;
4. Inhibition of neutrophil migration into joints and their associated release of degradative enzymes;
5. Fibrinolytic (anticoagulant) activity which improves blood circulation to bone and structures around the joint.

Glucosamine
Glucosamine is utilised in the production of glycosaminoglycans (GAGs), including hyaluronic acid (HA), and multiples of these complexes combine to form large proteoglycans in the matrix of cartilage. As such, glucosamine is a fundamental building block of the cartilage structure, and a growing body of evidence supports the pain-relieving and potentially chondroprotective properties of glucosamine. N-Acetyl Glucosamine actively stimulates and provides substrate for the secretion of GAGs in articular cartilage, as well as exerting an anti-inflammatory effect and inhibiting degradation of the connective tissues of the joints.1

Intravenous Administration - A multi-dose safety study in horses has demonstrated the safety of Pentosan GOLD + Halo when administered intravenously. At normal dosage, mild induced elevation of aPTT (activated partial thromboplastin time - measure of coagulation) was returned to baseline levels by 8 hours after each weekly injection during the 6 week trial.2

Indications
For intravenous administration to aid in the treatment of non-infectious inflammatory joint disease in horses.

Dosage and Administration
For adult horses (500 kg) draw up entire contents of both vials (total volume 18 mL) into a single syringe and administer by intravenous injection using a 21 gauge needle (or smaller).
This dose contains 1,500 mg Pentosan Polysulfate (3 mg/kg) + 2,400 mg N-Acetyl Glucosamine (4.8 mg/kg)

Warning
The efficacy and safety of this product combination has not been assessed by the APVMA. Contraindicated in horses with clotting defects, traumatic haemorrhage, infection, liver/kidney failure, or within 2 days of surgery.
